BCMA-CD24 CAR-T is an experimental bispecific chimeric antigen receptor (CAR) T-cell therapy developed for the treatment of multiple myeloma (MM). It is designed with an IF/THEN logic-gated circuit to overcome the limitations of traditional BCMA-targeted therapies, such as relapse driven by CD24-positive myeloma stem cells, while minimizing off-tumor toxicity. The construct features a constitutively expressed CAR targeting B-cell maturation antigen (BCMA) and an inducible CAR targeting CD24. The expression of the CD24-CAR is driven by a nuclear factor of activated T cells (NFAT) promoter, which is only activated when the BCMA-CAR binds to its target on myeloma cells. This mechanism ensures that the CD24-targeted cytotoxic activity is restricted to the myeloma microenvironment, sparing healthy tissues that widely express CD24.
