BCMA-CD3 CrossMAb refers to a bispecific antibody architecture that simultaneously targets B-cell maturation antigen (BCMA) and the CD3 epsilon subunit of the T-cell receptor complex. Utilizing the CrossMAb technology—which involves the crossover of antibody domains to ensure correct light-chain association—this modality functions as a T-cell engager. It facilitates the recruitment and activation of cytotoxic T cells to specifically eliminate BCMA-expressing malignant plasma cells. ProMab Biotechnologies has developed humanized versions of this antibody (using the 4C8A clone), including univalent and bivalent CrossMAb designs. In preclinical studies, these antibodies demonstrated high affinity for BCMA-positive cells, induced significant IFN-gamma secretion, and showed potent dose-dependent cytotoxic activity in multiple myeloma xenograft models.
