BCMA-CS1 cCAR is an autologous chimeric antigen receptor T-cell therapy engineered to express a compound CAR comprising two complete CARs, one targeting B-cell maturation antigen (BCMA/CD269) and the other targeting CS1 (also known as SLAMF7/CD319), linked by a self-cleaving P2A peptide to enable independent surface expression of both receptors on the same T cell.[5][7][10] By simultaneously recognizing BCMA- and/or CS1-expressing multiple myeloma cells, BCMA-CS1 cCAR is designed to enhance cytotoxicity against heterogeneous myeloma populations and reduce antigen escape that can occur with single-target BCMA or CS1 CAR-T cells.[5][10] Preclinical studies have demonstrated potent and specific in vitro and in vivo anti-myeloma activity against myeloma cell lines and primary tumor samples expressing various combinations of BCMA and CS1, and the product is being clinically developed for relapsed or refractory multiple myeloma.[5][7][10]
