Overview

BCMA-CS1 HLE BiTE is a dual-targeting bispecific T-cell engager (BiTE) developed by Amgen for the treatment of multiple myeloma. This immunotherapy agent is designed to simultaneously engage two tumor-associated antigens (TAAs) on cancer cells—B-cell maturation antigen (BCMA) and CS1 (also known as SLAMF7)—while binding to CD3ε on T cells. By bridging these targets, the molecule redirects the patient's T cells to induce lysis of the tumor cells. The molecule also incorporates an Fc-based domain to provide half-life extension (HLE), allowing for less frequent dosing compared to first-generation BiTE molecules. This dual-targeting approach is intended to overcome challenges such as heterogeneous antigen expression and potential antigen loss that can limit the efficacy of therapies targeting BCMA alone.
