BCMA-FAB-CD3 ScFv-Fc is a humanized bispecific antibody candidate developed by ProMab Biotechnologies for the treatment of multiple myeloma. It features a unique "triple chain" architecture consisting of a Fab fragment targeting B-cell maturation antigen (BCMA), a single-chain variable fragment (ScFv) targeting CD3, and an Fc region to enhance stability and pharmacokinetics. The molecule acts as a T-cell engager, simultaneously binding to BCMA on malignant plasma cells and the CD3 complex on T cells, thereby inducing T-cell activation and directed cytotoxicity against the tumor cells. Preclinical data presented at AACR 2022 demonstrated high binding affinity (Kd ~0.3 nM) and potent anti-tumor activity in MM1S xenograft mouse models.
