BCMA-Fc is a recombinant fusion protein consisting of the extracellular domain of B-cell maturation antigen (BCMA) fused to the Fc region of an immunoglobulin. This fusion protein functions as a decoy receptor that competitively binds to BCMA ligands, specifically A Proliferation-Inducing Ligand (APRIL) and B-cell activating factor (BAFF), preventing them from binding to membrane-bound BCMA on malignant plasma cells in multiple myeloma. By blocking APRIL and BAFF from engaging with BCMA on myeloma cells, BCMA-Fc inhibits downstream signaling cascades including the NF-κB, PI3K/AKT, and MAPK pathways, which are critical for myeloma cell proliferation, survival, and differentiation. The fusion protein also blocks APRIL-induced secretion of chemokines, including MIP-1α and MIP-1β, as well as other pro-survival and angiogenic factors. In preclinical studies, BCMA-Fc demonstrated the ability to inhibit adhesion and migration of myeloma cells and suppress APRIL-induced responses. This therapeutic approach represents a ligand-blocking strategy for treating multiple myeloma by disrupting the bone marrow microenvironment signaling that supports myeloma cell growth.
