BCMA-mutIL-2 is a novel CAR enhancer, a recombinant fusion protein designed to augment the activity and persistence of Chimeric Antigen Receptor (CAR) T cells, particularly B cell maturation antigen (BCMA) CAR T cells, for the treatment of multiple myeloma. It comprises a B cell maturation antigen (BCMA) component fused to a low-affinity interleukin 2 (IL-2) molecule. This construct selectively induces IL-2 signaling in CAR T cells upon antigen-CAR binding, thereby enhancing T cell activation and antitumor activity while mitigating IL-2-associated toxicities. It has been shown to increase CAR T cell proliferation, clearance of tumor cells, and development of memory CAR T cells.
