Overview

BCMA-PD1-CART is an engineered chimeric antigen receptor (CAR) T-cell therapy being investigated for the treatment of relapsed or refractory multiple myeloma. These CAR-T cells are designed to target the B-cell maturation antigen (BCMA) expressed on the surface of myeloma cells. A unique feature of this therapy is the inclusion of a genetic modification that causes the T-cells to secrete a high-affinity mutant PD-1 Fc fusion protein. This secreted protein acts as a localized checkpoint inhibitor by binding to PD-L1 on tumor cells and blocking its interaction with PD-1 on T-cells. By neutralizing this immunosuppressive pathway within the tumor microenvironment, BCMA-PD1-CART cells aim to resist exhaustion and maintain superior cytotoxic activity and persistence compared to conventional BCMA-targeting CAR-T therapies.
