BCMA-targeted allogeneic CAR-T refers to a class of "off-the-shelf" cellular immunotherapies designed to treat various cancers, primarily hematological malignancies like multiple myeloma. Unlike autologous CAR-T therapies, which use a patient's own T cells, allogeneic CAR-T therapies utilize T cells from healthy donors. These donor T cells are genetically engineered to express a chimeric antigen receptor (CAR) that specifically recognizes and binds to B-cell maturation antigen (BCMA), a protein highly expressed on multiple myeloma cells and other plasma cells. Further genetic modifications, such as knocking out genes like NKG2A, TRAC, and B2M, are often employed to reduce the risk of graft-versus-host disease (GvHD) and host immune rejection, enhancing the therapy's safety and efficacy. This approach aims to provide a more readily available and potentially more consistent treatment option compared to patient-specific autologous CAR-T therapies. Companies like CARsgen Therapeutics and Allogene Therapeutics are actively developing therapies in this class.
