BCMA-targeted PD-1-silenced CAR-T cells (also referred to as PD-1KD BCMA CAR-T cells) are an investigational autologous chimeric antigen receptor (CAR) T-cell therapy being developed for the treatment of relapsed or refractory multiple myeloma (RRMM). The therapy consists of autologous T cells genetically engineered to express a CAR targeting B-cell maturation antigen (BCMA/TNFRSF17) along with an OX-40 costimulatory domain, and co-transduced with a short hairpin RNA (shRNA) cassette designed to silence/knock down the expression of programmed cell death protein 1 (PD-1). By downregulating PD-1, these CAR-T cells are protected from PD-1/PD-L1-mediated immunosuppression within the tumor microenvironment, which helps preserve a memory T-cell phenotype, reduce exhaustion, and enhance antitumor efficacy and persistence. The therapy has been evaluated in early-phase clinical trials in China, demonstrating preliminary safety and efficacy.
