BCMA1 CAR T-cell therapy is an experimental chimeric antigen receptor (CAR) T-cell therapy targeting the B-cell maturation antigen (BCMA), developed by Lentigen (a Miltenyi Biotec company) for the treatment of multiple myeloma. The construct features a fully human single-chain variable fragment (scFv) derived from a yeast display library, which is linked to a CD8 hinge and transmembrane domain, a 4-1BB co-stimulatory domain, and a CD3ζ signaling domain. In preclinical evaluations, BCMA1 demonstrated potent cytotoxicity against BCMA-expressing myeloma cell lines such as RPMI-8226 and MM1.S. However, compared to the related BCMA2 construct, BCMA1 exhibited lower proliferative capacity and reduced persistence, characterized by a significant decline in the CD4+ T-cell subset during chronic target exposure.
