Overview

BCMA2 CAR-T is an experimental chimeric antigen receptor (CAR) T-cell therapy targeting the B-cell maturation antigen (BCMA), primarily developed for the treatment of multiple myeloma. Developed by Lentigen (a Miltenyi Biotec company), the therapy utilizes a fully human single-chain variable fragment (scFv) specific for BCMA, derived from yeast display libraries. The CAR construct is delivered via a lentiviral vector and includes a CD8 hinge and transmembrane domain, a 4-1BB co-stimulatory domain, and a CD3ζ signaling domain. A specialized "armored" variant, BCMA2-TbnegCAR, co-expresses a dominant negative TGF-β receptor II (TGF-β RII) to provide functional resistance against the immunosuppressive effects of TGF-β within the tumor microenvironment, thereby improving T-cell persistence, proliferation, and cytotoxic potency in multiple myeloma models.
