BCX-3607 is a small molecule inhibitor of the tissue factor/factor VIIa (TF/FVIIa) complex, originally developed by BioCryst Pharmaceuticals. It functions as a noncovalent, reversible, and competitive inhibitor that physically blocks the active site of the FVIIa catalytic domain. By targeting the initiation of the extrinsic coagulation pathway, BCX-3607 was designed to provide antithrombotic and anti-inflammatory effects without the excessive bleeding risks associated with traditional anticoagulants. Preclinical research demonstrated its ability to reduce thrombus weight in deep vein thrombosis (DVT) models and attenuate inflammatory responses, such as interleukin-6 production, in models of endotoxemia. The compound was primarily explored for the treatment of acute thrombotic conditions, including unstable angina and disseminated intravascular coagulation (DIC), though development was ultimately discontinued.
