Overview

BCX9250 is a novel, orally administered small molecule inhibitor of activin receptor-like kinase-2 (ALK-2), also known as ACVR1. It was discovered and developed by BioCryst Pharmaceuticals for the treatment of fibrodysplasia ossificans progressiva (FOP), an ultra-rare and severely disabling genetic disorder characterized by heterotopic ossification—the abnormal formation of bone in soft tissues such as muscles, tendons, and ligaments. The ALK-2 enzyme is part of the normal signaling pathway for bone formation; activating mutations in the ALK-2 gene are necessary for FOP to occur. By selectively inhibiting ALK-2 activity, BCX9250 aims to prevent or reduce heterotopic ossification in FOP patients. In preclinical models and Phase 1 clinical trials in healthy volunteers, BCX9250 demonstrated potency against its target kinase with favorable safety and tolerability profiles[2][5][6][8]. The drug has received Orphan Drug Designation from the FDA and EMA PRIME eligibility[1][4][5].
