Overview

BCX9499 is an orally administered small molecule inhibitor of activin receptor-like kinase 2 (ALK2), developed as a potential treatment for fibrodysplasia ossificans progressiva (FOP), a rare and severely debilitating genetic disorder characterized by heterotopic ossification (abnormal bone formation in soft tissue)[1][3][5][7]. BCX9499 works by selectively inhibiting ALK2 kinase, blocking disease-causing signaling activated by pathogenic mutations in the ALK2 gene (ACVR1), which are central to FOP pathogenesis[1][3][5]. Preclinical studies have shown that ALK2 inhibitors, including BCX9499, can dramatically reduce heterotopic ossification in animal models[1][3]. BCX9499 is in the development pipeline at BioCryst Pharmaceuticals, with the goal of advancing to Phase 1 clinical trials for FOP[1][4][5].
