BDC-3042

BDC-3042 is a proprietary, first-in-class agonist monoclonal antibody developed by Bolt Biotherapeutics. It targets dectin-2 (C-type lectin domain family 6 member A, CLEC6A), an immune-activating pattern recognition receptor highly expressed on tumor-associated macrophages (TAMs) in the tumor microenvironment. By agonizing dectin-2, BDC-3042 reprograms TAMs from an immunosuppressive to an immunostimulatory phenotype, promoting pro-inflammatory cytokine secretion and enhancing antigen presentation. This leads to robust CD8+ T cell-mediated anti-tumor immunity and has shown biological activity with dose-dependent increases in pro-inflammatory markers and signs of anti-tumor efficacy in early clinical studies. The drug is being evaluated as monotherapy and in combination with checkpoint inhibitors for advanced solid tumors including non-small cell lung cancer, triple-negative breast cancer, renal cell carcinoma, colorectal cancer, melanoma, ovarian cancer, colon cancer, and head and neck cancers[1][3][4][5][7][9].
