Overview

BDC-4182 is a next-generation Boltbody ISAC (immune-stimulating antibody conjugate) developed by Bolt Biotherapeutics. It targets claudin 18.2, a clinically validated antigen expressed in gastric cancer, gastroesophageal junction cancer, pancreatic cancer, and other tumor types. The drug combines a monoclonal antibody specific for claudin 18.2 with a potent TLR7/8 agonist payload via optimized conjugation chemistry to enhance immune activation and phagocytosis of tumor cells. In preclinical models, BDC-4182 demonstrated significant anti-tumor activity—including in tumors with low claudin 18.2 expression—elicited immunological memory, and outperformed cytotoxic claudin 18.2 ADCs (antibody-drug conjugates). The safety profile was acceptable in non-human primates at the highest tested dose (12 mg/kg), supporting further clinical development with an initial trial expected to begin in 2025[1][2][4][5][6].
