BE CAR-7 (also known as TvT CAR7) is an investigational, allogeneic (universal) CAR-T cell therapy targeting CD7, developed by University College London (UCL) and Great Ormond Street Hospital (GOSH). It is specifically designed to treat relapsed or refractory T-cell acute lymphoblastic leukemia (T-ALL), a condition where traditional autologous CAR-T approaches are hampered by the risk of "fratricide" (CAR-T cells killing each other) and the difficulty of harvesting healthy T cells from the patient. BE CAR-7 utilizes CRISPR cytosine base editing to perform a triple gene knockout: *CD7* (to prevent fratricide), the T-cell receptor (specifically *TRBC* or *TRAC* to prevent graft-versus-host disease), and *CD52* (to permit the use of alemtuzumab for selective lymphodepletion). This "off-the-shelf" product is derived from healthy donors and is intended to induce deep remission, serving as a bridge to a consolidative stem cell transplant.
