Drug intelligence / Profile preview
BEA-28 is a first-in-class, cereblon-recruiting small molecule targeted protein degrader (PROTAC) that degrades TEAD (TEA Domain) transcription factors, which are key effectors of the Hippo-YAP/TAZ signaling pathway. Developed by Beactica Therapeutics using its Eclipsor platform, BEA-28 is designed to remove TEAD proteins and all their functions rather than simply blocking a binding pocket. This mechanism is particularly suited for treating cancers where Hippo pathway dysregulation drives tumor growth, immune evasion, or resistance to targeted therapies. In preclinical studies, BEA-28 has demonstrated robust tumor regression in xenograft models of mesothelioma and KRAS-mutant lung cancer, and has shown the ability to reverse acquired resistance to KRAS inhibitors in cellular models. Beactica is advancing BEA-28 as a preclinical candidate with plans for biomarker-selected combination strategies involving checkpoint inhibitors and KRAS inhibitors.
