BEAM-103 is an investigational monoclonal antibody targeting CD117 (also known as c-Kit), a receptor tyrosine kinase expressed on the surface of hematopoietic stem and progenitor cells (HSPCs). Developed as part of Beam Therapeutics' ESCAPE (Engineered Stem Cell Antibody Evasion) platform, BEAM-103 is designed to selectively deplete host HSPCs to create space for engraftment during stem cell transplantation. Unlike traditional chemotherapy or radiation-based conditioning regimens, BEAM-103 is intended to serve as a non-genotoxic conditioning agent, potentially reducing the severe toxicities associated with myeloablative therapy. It is specifically being developed for use in a therapeutic regimen for sickle cell disease, where it is intended to be paired with BEAM-104, a base-edited autologous cell therapy engineered to be resistant to BEAM-103-mediated depletion.
