Overview

BEBT-209 is an investigational, orally administered small molecule inhibitor targeting cyclin-dependent kinases 4 and 6 (CDK4/6). It is being developed primarily for the treatment of advanced solid tumors, with a focus on breast cancer subtypes such as hormone receptor-positive/HER2-negative breast cancer and advanced triple-negative breast cancer (TNBC). The drug acts by inhibiting CDK4 and CDK6, key regulators of cell cycle progression from G1 to S phase, thereby blocking tumor cell proliferation. Clinical studies have shown that BEBT-209 in combination with chemotherapy agents like gemcitabine and carboplatin demonstrates promising efficacy and manageable toxicity in metastatic TNBC patients. The most common adverse events are hematologic toxicities consistent with the chemotherapy backbone. BEBT-209 is currently in Phase 2/3 clinical trials for various forms of advanced breast cancer[2][4][5][6][10].
