BEBT-209 + carboplatin + gemcitabine is a combination regimen under clinical investigation for advanced and metastatic triple-negative breast cancer (TNBC). BEBT-209 is a potent, selective cyclin-dependent kinase 4 (CDK4) inhibitor; it inhibits CDK4 to block cell cycle progression in cancer cells, potentially enhancing antitumor activity when combined with cytotoxic chemotherapy agents. Carboplatin is a platinum-based DNA-damaging agent, and gemcitabine is a nucleoside analog that interferes with DNA synthesis. Together, this combination aims to leverage cell cycle arrest (via BEBT-209) and enhanced cytotoxicity from carboplatin and gemcitabine. Early-phase clinical trials show encouraging efficacy with manageable toxicity, notably in TNBC patients with early relapse or post-systemic therapy.
