BECC44 is a structurally modified Toll-like receptor 4 (TLR4) agonist developed through bacterial enzymatic combinatorial chemistry (BECC). It is an engineered derivative of lipopolysaccharide (LPS) designed to maintain the potent antitumor immune-activating properties of endotoxins—specifically the induction of TNF⍺ and IFNγ—while significantly reducing the systemic toxicity and sepsis-like symptoms associated with native LPS. In preclinical studies of pancreatic ductal adenocarcinoma (PDAC), BECC44 has shown efficacy in reducing tumor burden by stimulating cytotoxic CD4⁺ T-cell-mediated immunity. It represents a next-generation immunotherapeutic approach aimed at overcoming the immunosuppressive environment of pancreatic tumors.
