Overview

Becotatug vedotin + cisplatin is a combination therapy consisting of becotatug vedotin (MRG003), an antibody-drug conjugate (ADC) targeting the epidermal growth factor receptor (EGFR), and cisplatin, a platinum-based chemotherapeutic agent. Becotatug vedotin utilizes a humanized anti-EGFR monoclonal antibody conjugated to the microtubule-disrupting agent monomethyl auristatin E (MMAE) via a valine-citrulline linker. Cisplatin acts by forming DNA cross-links, leading to apoptosis. This combination is primarily being investigated for the treatment of locally advanced esophageal squamous cell carcinoma (ESCC), often in conjunction with radiotherapy, to leverage the targeted delivery of cytotoxic agents alongside systemic DNA-damaging chemotherapy.
