Bederocin (REP8839) is a novel diaryldiamine-containing small molecule antibiotic developed as a topical agent for the treatment of skin and wound infections. Its mechanism of action is the selective inhibition of methionyl-tRNA synthetase (MetRS), an essential enzyme in bacterial protein synthesis. This inhibition leads to potent antibacterial activity against a range of gram-positive pathogens, including *Staphylococcus aureus* (notably methicillin-resistant *S. aureus*, MRSA), *Staphylococcus epidermidis*, *Streptococcus pyogenes*, *Enterococcus faecium*, and *Enterococcus faecalis*—even strains resistant to other antibiotics such as vancomycin and mupirocin. REP8839 demonstrates high specificity for bacterial MetRS with minimal activity against mammalian counterparts, making it suitable for topical use with low systemic absorption. The drug was originally developed by Replidyne and has undergone Phase I clinical trials demonstrating safety and tolerability in humans[1][3][4][5].
