Overview

BeEAM is a high-dose chemotherapy conditioning regimen consisting of bendamustine, etoposide, cytarabine, and melphalan. It was developed by the Centre Léon Bérard as an alternative to the standard BEAM (carmustine, etoposide, cytarabine, and melphalan) regimen, specifically replacing carmustine with bendamustine. The regimen was intended for use prior to autologous stem cell transplantation (ASCT) in patients with first or second chemosensitive relapsed follicular lymphoma. However, a Phase II multicenter study was terminated prematurely due to unacceptable toxicities, including severe infectious complications and veno-occlusive disease. The regimen functions through multiple cytotoxic mechanisms, including DNA alkylation and inhibition of DNA synthesis and repair.
