Belagenpumatucel-l is a therapeutic cancer vaccine developed for the treatment of non-small cell lung cancer (NSCLC). It is prepared by transfecting allogeneic NSCLC cells with a plasmid containing an antisense transgene targeting transforming growth factor beta 2 (TGF-beta2), followed by expansion, irradiation, and freezing of the cells. Upon administration, this vaccine aims to elicit a cytotoxic T lymphocyte response against host NSCLC cells and suppress tumor-derived TGF-beta2 production, thereby enhancing immunogenicity and reducing tumor proliferation. The drug was primarily developed as maintenance therapy for advanced NSCLC but has also been investigated in glioma. Its mechanism involves both immunostimulation and inhibition of transforming growth factor beta signaling[1][3][5][8].
