Overview

Belinostat is a small molecule histone deacetylase (HDAC) inhibitor used for the treatment of relapsed or refractory peripheral T-cell lymphoma (PTCL). It inhibits HDAC enzymes, leading to the accumulation of acetylated histones and other proteins, which induces cell cycle arrest and/or apoptosis in transformed cells. Belinostat shows preferential cytotoxicity toward tumor cells compared to normal cells. The drug was developed by TopoTarget and is administered intravenously as monotherapy, typically on days 1–5 of a 21-day cycle. It received FDA approval in July 2014 for PTCL after other treatments have failed.
