Overview

Belinostat + CHOP is a combination chemotherapy regimen consisting of belinostat, a pan-histone deacetylase inhibitor, and the standard CHOP regimen. CHOP comprises cyclophosphamide, doxorubicin, vincristine sulfate, and prednisone. Belinostat exerts antineoplastic activity by inhibiting histone deacetylase enzymes, resulting in altered gene expression and apoptosis in malignant cells. The combination is under investigation as a first-line treatment for peripheral T-cell lymphoma (PTCL), aiming to improve the poor outcomes typically seen with CHOP alone. Clinical studies show that the regimen is generally well-tolerated, with manageable toxicities and a high rate of complete and partial responses in newly diagnosed PTCL patients[1][2][3][4].
