Bellamine S is a fixed-dose combination pharmaceutical product consisting of belladonna alkaloids, ergotamine tartrate, and phenobarbital, formulated as an oral extended-release tablet. It was historically utilized to manage a variety of conditions related to autonomic nervous system imbalance, most notably menopausal symptoms such as hot flashes, night sweats, and restlessness. The therapeutic effect is achieved through the synergistic actions of its three components: belladonna alkaloids provide anticholinergic activity to reduce glandular secretions and smooth muscle spasms; ergotamine tartrate acts as a vasoconstrictor on cranial blood vessels to treat vascular headaches; and phenobarbital serves as a sedative-hypnotic by enhancing GABAergic inhibition in the central nervous system. Due to the inclusion of a barbiturate, the drug carries risks of dependency and significant drug-drug interactions. Most formulations of this type have been discontinued or superseded by safer alternatives in modern clinical practice.
