Overview

This is an investigational **combination therapy** comprising three agents: - **Belrestotug**: a fully human IgG1 monoclonal antibody targeting the immunoregulatory receptor TIGIT on lymphocytes, acting primarily as a TIGIT antagonist. It enhances anti-tumor immune response via inhibition of TIGIT-mediated immune suppression and promotion of T lymphocyte activity, with additional mechanisms via antibody-dependent cell cytotoxicity. Developed initially by iTeos Therapeutics and subsequently in partnership with GSK, belrestotug reached Phase II/III clinical development in several cancers (notably non-small cell lung cancer and solid tumors)[1][3][4][5][7][9]. - **Iberdomide**: a novel oral cereblon E3 ligase modulator (CELMoD). Iberdomide modulates immunomodulatory and anti-proliferative effects through targeted protein degradation, resulting in downregulation of Ikaros and Aiolos transcription factors—central to cell survival in multiple myeloma and other hematologic malignancies. It is classified as a small molecule immunomodulator. - **Dexamethasone**: a synthetic glucocorticoid corticosteroid commonly used as an adjunct in hematological malignancies due to its anti-inflammatory, lympholytic, and anti-tumor effects, mainly through broad immunosuppression and induction of apoptosis in lymphoid cells. This combination is studied primarily in hematologic malignancies, especially **multiple myeloma**, where layered immunomodulation and immune checkpoint blockade may synergistically target resistant and relapsed disease.
