Bemarituzumab + paclitaxel + ramucirumab is an investigational combination therapy being studied primarily for advanced or metastatic adenocarcinoma of the stomach or gastroesophageal junction, particularly in patients who have failed prior chemotherapy. Bemarituzumab is a monoclonal antibody targeting Fibroblast growth factor receptor 2b (FGFR2b), aiming to inhibit tumor growth in cancers overexpressing this receptor[1][5]. Paclitaxel is a well-established small molecule chemotherapeutic agent that stabilizes microtubules and inhibits cell division. Ramucirumab (brand name Cyramza) is a human IgG1 monoclonal antibody that binds to Vascular endothelial growth factor receptor 2 (VEGFR2), blocking its activation by VEGF ligands and thereby inhibiting angiogenesis critical for tumor progression[3][6]. The combination leverages anti-tumor activity through both direct cytotoxicity (paclitaxel), inhibition of angiogenesis (ramucirumab), and targeted blockade of Fibroblast growth factor receptor 2b (FGFR2b) signaling pathways (bemarituzumab).
