Bemotuzumab (HLX20) is a humanized monoclonal antibody of the IgG1-kappa isotype that targets the **programmed death-ligand 1 (PD-L1)**. Developed by Shanghai Henlius Biotech, it functions as an immune checkpoint inhibitor. By binding specifically to PD-L1 on the surface of tumor cells and tumor-infiltrating immune cells, bemotuzumab prevents its interaction with the PD-1 and B7.1 receptors on T cells. This blockade interrupts the inhibitory signaling pathway that allows tumors to evade immune detection, thereby reactivating the anti-tumor activity of cytotoxic T lymphocytes. Bemotuzumab is currently undergoing clinical evaluation in several indications, most notably as part of neoadjuvant chemotherapy regimens for triple-negative breast cancer (TNBC) to enhance pathological complete response (pCR) rates, and in early-phase trials for advanced solid tumors.
