Bempegaldesleukin (NKTR-214) is an investigational immunotherapy designed as a CD122-preferential interleukin-2 (IL-2) pathway agonist. It is engineered to stimulate the proliferation and activation of tumor-infiltrating lymphocytes (TILs), particularly CD8+ T cells and natural killer (NK) cells within the tumor microenvironment. Nivolumab is a monoclonal antibody targeting programmed cell death protein 1 (PD-1), blocking its interaction with PD-L1/PD-L2 to enhance T-cell mediated immune responses against tumors. Ipilimumab is a monoclonal antibody that targets cytotoxic T lymphocyte-associated antigen 4 (CTLA-4), promoting T-cell activation and proliferation by inhibiting this immune checkpoint. The combination of these three agents aims to synergistically enhance anti-tumor immunity through complementary mechanisms—expanding effector immune cells while simultaneously releasing inhibitory checkpoints on their activity. This triplet regimen has been investigated in advanced solid tumors including melanoma, renal cell carcinoma, non-small cell lung cancer, and urothelial carcinoma[4][5].
