BEN8744 is a potent, peripherally restricted, orally administered small molecule inhibitor of phosphodiesterase 10 (PDE10), developed as a potential first-in-class treatment for ulcerative colitis and other inflammatory bowel diseases. Unlike previous PDE10 inhibitors that caused central nervous system (CNS) side effects due to brain penetration, BEN8744 has been specifically designed to minimize CNS exposure and associated adverse events. Preclinical studies demonstrated robust anti-inflammatory activity in patient-derived colonic biopsies from both ulcerative colitis and Crohn’s disease patients, with reductions in pro-inflammatory cytokines such as IL6, IL8, and TNFα comparable to standard-of-care agents like JAK inhibitors and steroids. The drug has completed Phase 1 clinical trials showing favorable safety, tolerability, pharmacokinetics, and no evidence of CNS-related side effects at any dose level.
