Overview

Benchmark CLL1 CAR-T refers to a specific chimeric antigen receptor (CAR) T-cell therapy construct targeting C-type lectin-like molecule 1 (CLL1, also known as CLEC12A), which is utilized as a reference standard in comparative studies for myeloid malignancies. The construct is defined by the molecular architecture and sequences disclosed in patent WO2017091615A, originally developed by researchers at Massachusetts General Hospital and the Broad Institute. It is designed to target CLL1, a type II transmembrane glycoprotein highly expressed on acute myeloid leukemia (AML) blasts and leukemic stem cells but absent on normal hematopoietic stem cells, thereby providing a tumor-specific target. In preclinical and clinical development contexts, such as those reported by Fundamenta Therapeutics at ASCO 2024, this benchmark is used to evaluate the relative potency, expansion, and safety of novel CAR-T platforms like the allogeneic ThisCART system.
