Benchmark PRAME TCR-T refers to established or clinical-stage T-cell receptor (TCR) engineered T-cell therapies targeting the Preferentially Expressed Antigen in Melanoma (PRAME). These therapies serve as reference standards in comparative studies for next-generation TCR-T candidates. PRAME is a cancer-testis antigen highly expressed in various solid tumors, including melanoma, sarcoma, and lung cancer, while having limited expression in normal tissues, making it an ideal target for immunotherapy. Benchmark products typically utilize a high-affinity TCR to recognize PRAME-derived peptides presented by HLA-A*02:01, though they may lack the advanced modifications (such as switch receptors or co-receptors) found in newer supercharged iterations.
