Bendamustine + ofatumumab is a combination therapy regimen consisting of bendamustine, a bifunctional alkylating agent with unique cytotoxicity properties, and ofatumumab, an anti-CD20 monoclonal antibody. Bendamustine causes DNA damage through alkylation and cross-linking, while ofatumumab binds to CD20 on B lymphocytes to induce complement-dependent cytotoxicity and antibody-dependent cellular cytotoxicity. This combination has been studied for the treatment of various B-cell malignancies including chronic lymphocytic leukemia, mantle cell lymphoma, and indolent B-cell non-Hodgkin lymphoma.
