This combination regimen is an adaptive chemo-immunotherapy treatment being evaluated for treatment-naïve mantle cell lymphoma (MCL). It consists of **bendamustine**, a nitrogen mustard alkylating agent that induces DNA cross-linking; **orelabrutinib**, a potent and highly selective irreversible inhibitor of Bruton's tyrosine kinase (BTK); and **zuberitamab** (HS006), a recombinant human-mouse chimeric anti-CD20 monoclonal antibody. The regimen is the focus of the ADAPT-MCL Phase II study (NCT05492500) led by Tianjin Medical University Cancer Institute and Hospital. This study utilizes an adaptive design where treatment is tailored based on the patient's Minimal Residual Disease (MRD) and PET-CT status, potentially allowing patients who achieve a complete molecular response to transition from chemotherapy-based induction to a targeted maintenance phase.
