Bendamustine + rituximab + ibrutinib (IBR) is a combination therapy regimen that combines two chemotherapeutic agents with a targeted therapy. This regimen leverages complementary mechanisms of action: - Bendamustine is an alkylating agent that damages DNA, resulting in cancer cell death - Rituximab is a monoclonal antibody targeting CD20 on B cells - Ibrutinib is a Bruton's tyrosine kinase (BTK) inhibitor that blocks B-cell receptor signaling The addition of ibrutinib to the bendamustine + rituximab (BR) backbone has shown significant improvements in progression-free survival in patients with chronic lymphocytic leukemia (CLL) and certain types of non-Hodgkin lymphoma. In clinical trials, this combination has demonstrated efficacy in both previously untreated patients and those with relapsed/refractory disease.
