This combination therapy consists of benidipine hydrochloride, a dihydropyridine calcium channel blocker with affinity for T-type and L-type channels, and an unspecified beta-adrenergic antagonist. It was evaluated in the Phase 4 COPE (Combination Therapy of Hypertension to Prevent Cardiovascular Events) trial, an investigator-initiated multicenter study led by Dr. Seiji Umemoto at Yamaguchi University in collaboration with Kyowa Hakko Kirin. The trial compared the efficacy and safety of three benidipine-based combination regimens in Japanese hypertensive patients to determine which best prevents cardiovascular events. Findings from the study suggested that the benidipine and beta-blocker combination was less effective in reducing the risk of stroke and other hard cardiovascular endpoints compared to combinations involving thiazide diuretics.
