Overview

Benorilate (also known as benorylate) is an ester-linked codrug formed from acetylsalicylic acid (aspirin) and paracetamol (acetaminophen). It acts as an anti-inflammatory, analgesic, and antipyretic agent. After oral administration, it is absorbed and rapidly hydrolyzed to release its active components—salicylate and paracetamol—which then exert their usual pharmacological effects. The primary mechanism of action involves inhibition of cyclooxygenase (COX) enzyme, leading to reduced synthesis of prostaglandins that mediate pain, fever, and inflammation. Benorilate has been used for the treatment of mild to moderate pain, fever, osteoarthritis, rheumatoid arthritis, and soft-tissue rheumatism. Compared to aspirin alone or in combination with paracetamol taken separately, it may have improved gastric tolerance but is less effective for childhood fever; due to conversion into aspirin in vivo it should not be used in children because of the risk of Reye syndrome[2][3][5][6].
