Overview
Benralizumab is a humanized, afucosylated IgG1k monoclonal antibody that targets the alpha subunit of the interleukin-5 receptor (IL-5Rα) on eosinophils and basophils. By binding to IL-5Rα, it blocks IL-5 signaling and induces apoptosis of these cells through enhanced antibody-dependent cell-mediated cytotoxicity (ADCC), primarily via natural killer (NK) cells. This leads to a significant reduction in eosinophil counts in blood and tissues. Benralizumab is used as an add-on maintenance treatment for severe eosinophilic asthma in patients aged 6 years or older whose disease is not controlled with current therapies, and for adult patients with eosinophilic granulomatosis with polyangiitis (EGPA). It was developed by MedImmune/AstraZeneca and approved by the FDA in 2017[1][2][3][4][5][6].
