A combination of two monoclonal antibodies used for the treatment of severe asthma and other type 2 inflammatory conditions. Benralizumab is a humanized recombinant monoclonal antibody that binds to the alpha chain of the interleukin-5 receptor (IL-5Rα) expressed on eosinophils and basophils. It works through a unique mechanism involving antibody-dependent cell-mediated cytotoxicity (ADCC), which leads to the depletion of eosinophils in circulation, bone marrow, and target tissues. Dupilumab is a monoclonal antibody that targets the interleukin-4 receptor alpha (IL-4Rα), blocking both IL-4 and IL-13 signaling pathways. This dual biologic approach is used in cases where a single biologic fails to effectively manage severe asthma, particularly in patients with comorbid conditions such as atopic dermatitis, chronic rhinosinusitis with nasal polyps, or eosinophilic esophagitis.
