Overview

Benufutamab (GEN1029) is a first-in-class bispecific monoclonal antibody mixture developed using the HexaBody technology. It consists of two noncompeting IgG1 antibodies targeting distinct epitopes on death receptor 5 (DR5; also known as TNFRSF10B or TRAILR2), each engineered with an E430G mutation in the Fc domain to enhance hexamerization. This design enables potent DR5 clustering and activation independent of Fcγ receptor engagement, leading to efficient induction of apoptosis in tumor cells. Benufutamab was developed for the treatment of various solid tumors and has demonstrated strong preclinical antitumor activity across multiple cancer types[1][4][8]. The drug was originally discovered by iDD Biotech and further developed by Genmab.
