Drug intelligence / Profile preview
Lentocilin S 1200 is a long-acting formulation of benzathine benzylpenicillin (penicillin G benzathine) developed and manufactured by Laboratorios Atral in Portugal. It is supplied as a powder for reconstitution with a co-packaged 1.5% lidocaine hydrochloride diluent to form a suspension for deep intramuscular injection. As a beta-lactam antibiotic, benzathine benzylpenicillin acts by binding to and inhibiting bacterial penicillin-binding proteins (PBPs), which are essential enzymes involved in the final transpeptidation step of peptidoglycan cell wall synthesis. This inhibition compromises cell wall integrity, leading to osmotic instability and bacterial cell lysis. Lentocilin S 1200 is indicated for the treatment of various bacterial infections, including group A streptococcal upper respiratory tract infections, syphilis (primary, secondary, latent, tertiary, and congenital), yaws, bejel, and pinta, as well as for the prophylaxis of rheumatic fever and diphtheria. Due to global shortages of Bicillin L-A, regulatory agencies such as Health Canada and the US FDA have authorized the temporary exceptional importation and distribution of Lentocilin S 1200 (imported by Septa Pharmaceuticals in Canada and distributed by Mark Cuban Cost Plus Drug Company in the US) to maintain the supply of this critical therapeutic.
