Overview

Benzydamine is a non-steroidal anti-inflammatory drug (NSAID) with local anesthetic and analgesic properties, primarily used for the treatment of pain and inflammation in the mouth and throat. It is typically applied topically as a mouthwash, spray, lozenge, or cream. Benzydamine acts mainly by inhibiting the synthesis of pro-inflammatory cytokines such as tumor necrosis factor-alpha (TNF-α) and interleukin-1β (IL-1β), with minimal effect on other cytokines or anti-inflammatory mediators. Unlike traditional NSAIDs that inhibit cyclooxygenase enzymes directly, its anti-inflammatory action is related to inhibition of the p38 MAPK pathway and chemotactic cell migration. Benzydamine accumulates in inflamed tissues due to its basic character and has limited systemic absorption when used topically[1][4][5][8]. It was first synthesized in Italy in 1964.
