Benzylthiouracil is a small molecule antithyroid drug of the thioamide class, closely related to propylthiouracil. It acts by inhibiting thyroid peroxidase, thereby blocking the synthesis of thyroid hormones (T3 and T4) but not their release. This leads to a reduction in circulating thyroid hormone levels once preformed stores are depleted. Benzylthiouracil is primarily indicated for the treatment of hyperthyroidism and multinodular goiter. The drug has been associated with severe adverse effects, including vasculitis (notably ANCA-positive glomerulonephritis), toxic hepatitis, and lung damage[1][4][5].
