Overview

Beperminogene perplasmid (AMG0001) is a non-viral, plasmid DNA-based gene therapy developed by AnGes for the treatment of critical limb ischemia (CLI) and peripheral arterial disease. It consists of a circular DNA plasmid (pVAX1) encoding the human hepatocyte growth factor (HGF) gene. Administered via local intramuscular injection, the plasmid is taken up by myocytes, which then express and secrete HGF protein. HGF is a potent angiogenic factor that binds to the c-Met receptor on endothelial and smooth muscle cells, triggering signaling pathways that promote the formation of new collateral blood vessels (angiogenesis). This mechanism improves blood perfusion to ischemic tissues, facilitates the healing of peripheral ischemic ulcers, and reduces rest pain. It received conditional and term-limited marketing approval in Japan under the brand name Collategene, specifically for CLI patients with Buerger's disease or peripheral arterial disease who have limited options for surgical revascularization.
