Overview
Bepirovirsen is an investigational antisense oligonucleotide (ASO) designed for the treatment of chronic hepatitis B virus (HBV) infection. It works by specifically recognizing and binding to HBV RNA in infected liver cells (hepatocytes), leading to degradation of the viral RNA and a reduction in both viral replication and production of hepatitis B surface antigen (HBsAg). This triple-action mechanism includes inhibition of viral DNA replication, suppression of HBsAg levels in the blood, and stimulation of immune responses—potentially via Toll-like receptor 8 activation—to increase the chances of achieving a durable functional cure. Bepirovirsen is currently being evaluated in phase III clinical trials as both monotherapy and as part of sequential regimens with nucleoside/nucleotide analogues. The drug was discovered by Ionis Pharmaceuticals and further developed through collaboration with GSK[1][2][4][6][7].
