Overview
**Bepirovirsen** is a first-in-class, unconjugated 2'-O-methoxyethyl-modified antisense oligonucleotide developed by Ionis Pharmaceuticals and licensed to GSK for chronic hepatitis B. It hybridizes to a conserved sequence shared across hepatitis B virus RNA transcripts, including messenger RNAs and pregenomic RNA, promoting RNase H1-mediated degradation of viral RNA. This reduces viral protein production, including hepatitis B surface antigen, and suppresses viral replication, supporting restoration of immune control. It is administered by subcutaneous injection and is marketed in the United States as Hibsago for selected adults with chronic hepatitis B receiving nucleos(t)ide analogue therapy.
